Contichrom® TWIN 500 HPLC
The Contichrom® TWIN 500 HPLC is an industrial high-pressure chromatography system engineered for commercial-scale manufacturing of synthetic peptides, oligonucleotides, and small molecules. Operating at 80 bar with proprietary YMC fast-acting valves — the only valve technology capable of rapid switching at high pressure — the TWIN HPLC enables MCSGP continuous polishing with AutoPeak® to eliminate the yield-purity trade-off. Achieve +10–50% absolute yield increase at target purity and up to 70% less solvent consumption compared to batch HPLC. Class I Division 2 certified (ATEX Zone 2 optional) for safe operation with organic solvents, with SS 316L or Hastelloy flow paths for reversed-phase and ion exchange chromatography. The TWIN 500 has flow rates from 0.08 – 8.33 L/min.

| Flow Path | Best For | Reason |
|---|---|---|
| SS 316L | Reversed phase (RP) | Compatible with organic solvents (ACN, MeOH) |
| Hastelloy | Ion exchange (AIEX/CIEX) | Corrosion-resistant for high-salt, acid, and base eluents |
Specify your flow path material at time of order based on your primary chromatography mode.
YMC fast acting valves are the only fast-acting valve technology capable of reliable switching at 80 bar. This is critical for MCSGP, where column interconnection and side-fraction routing must happen rapidly during elution to maintain separation quality. Standard pneumatic valves cannot switch fast enough at these pressures.

- N-Rich® with AutoPeak® — Impurity enrichment
- 2D Integrated Batch — Multi-step automated purification
- Batch — Single-column gradient and isocratic operation
- All accessible from a single platform at 80 bar
- YMC valves enable rapid switching that makes MCSGP possible at high pressure
MCSGP with AutoPeak® — Continuous Polishing
Twin-column continuous polishing that eliminates the purity-yield trade-off:
- Impure side-fractions automatically identified and recycled to the second column
- Near-complete recovery at target purity — no re-chromatography
- AutoPeak® dynamically adjusts collection windows via real-time UV
- YMC valves switch rapidly during elution at full 80-bar pressure
- Continuous, automated operation for 24/7 manufacturing
Target: Gradient polishing separations (RP, IEX) Molecules: Synthetic peptides, oligonucleotides, small molecules, nutraceuticals
MCSGP — Performance Gains
| Metric | Result |
|---|---|
| Yield | 30–60% higher (up to 3× vs. batch) |
| Solvent consumption | Up to 70% reduction |
| Re-chromatography | Eliminated |
| Column size | Shorter columns — improved productivity |
| Synthesis batch size | Downscaling possible via higher yields |
| Process control | AutoPeak® enables continuous 24/7 operation |
2D Integrated Batch — Multi-Step Automation
Couple two orthogonal chromatography steps in a single automated process:
- Product from column 1 transfers directly to column 2 with in-line dilution
- No intermediate hold tanks or manual handling
- Example: RP polishing (Dim 1) → AIEX polishing (Dim 2)
- Reduces process time, QC burden, and facility footprint
Molecules: Synthetic peptides, oligonucleotides, small molecules, nutraceuticals
N-Rich with AutoPeak® — Impurity Enrichment
Automated twin-column cyclic process for isolating low-abundance compounds:
- Fresh feed added each cycle while depleting unwanted products
- Target compounds continuously recycled and accumulated on-column
- Up to 80× faster than traditional pooled HPLC approaches
- AutoPeak® compensates for retention shifts across many cycles
- Drastically reduces manual handling and project time
Target Molecules: Low-abundance compounds
N-Rich — Performance Gains
| Metric | Result |
|---|---|
| Processing speed | Up to 80× faster than pooled HPLC |
| Manual handling | Drastically reduced |
| Purity | Significantly higher than semi-prep batch |
| Project time | Dramatically shortened |
| Process control | AutoPeak® ensures consistency across all cycles |
Four stages: Design → Enrichment → Depletion → Elution.

| Model | Column ID | Best For |

| TWIN 500 | 20–45 cm | Mid-scale commercial production |
| TWIN 1000 | 30–60 cm | Large-scale commercial production |
