How it works

Twin-Column Hardware Built for Continuous Chromatography

The Contichrom® CUBE is a compact, two-module system that stacks into a 120 × 70 cm benchtop footprint. Two column positions are plumbed in parallel, enabling both interconnected and independent operation — the foundation of all continuous processes. Every component is engineered for the automated, high-pressure switching sequences that power CaptureSMB®, MCSGP, and N-Rich®.

Four High-Precision Pumps

Pump Role Location
P1A + P1B Binary gradient pump CUBE+
P2 Isocratic / Feed pump CUBE
P3 Isocratic / Wash pump CUBE

All pumps are rated to 100 bar with ±2% accuracy, 0.5% RSD precision, and active seal wash to prevent salt precipitation during long continuous runs.

16-Inlet Buffer & Sample Selection

Pumps P2 and P3 are each equipped with an 8-port selection valve, providing 16 independently addressable inlets for automated switching between buffers, wash solutions, and feed streams. Combined with the binary gradient pump (P1A/P1B), the system supports complex multi-step methods without manual intervention.

Intelligent Valve Network

Six multi-position rotary valves and two drain valves form the system’s fluidic “traffic controller.” Four column valves (V1A, V1B, V2A, V2B) direct flow to, from, and between both columns — executing the automated switching sequences that define each continuous process.

Dual UV Detectors

One detector sits at the outlet of each column — critical for dynamic process control.

  • Type: multi-wavelength, 4-channel Diode Array (DAD)
  • Range: 200–600 nm
  • Path length: 0.5 mm (exchangable for 0.1, 0.3, & 1 mm options)
  • Linear range: 0–1000 mAU

These detectors provide the real-time UV signals that enable AutoPeak® and AutomAb® dynamic process control.

In-Line Conductivity & pH

Two conductivity sensors (0–250 mS/cm) are positioned after each UV detector for accurate gradient monitoring. An external pH sensor (range 1–14) at the product outlet completes the in-line analytical suite — giving full real-time visibility of your separation.

Dynamic Mixer

Installed after the gradient pumps (P1A/P1B), the dynamic mixer uses a magnetic stirrer chamber to ensure homogeneous mobile phase composition. Essential for reproducible gradients in reversed-phase and mixed-mode applications.

  • Volume: 1.1 mL standard, 5.9 mL with upper chamber

Automated Fraction Collection

The system supports Teledyne ISCO Foxy R1 (single rack) and R2 (dual rack) fraction collectors for fully automated, walk-away operation.

  • Formats: 96-deep-well plates, 15 mL & 50 mL tubes, 1.5 mL microcentrifuge tubes
  • Protection: Diverter valve with drip tray prevents cross-contamination

Biocompatible Flow Path

Every wetted surface is selected for chemical resistance and biocompatibility.

Component Wetted Materials
Pumps PEEK, Ruby, Sapphire, Zirconia, FFKM
Valves PEEK, PPS, PTFE composite
Tubing PEEK (high-pressure), FEP (low-pressure)
Detectors PEEK, Quartz, PTFE, Stainless Steel

High-pressure tubing: 0.75 mm ID (CUBE 30) or 1.0 mm ID (CUBE 100).

Process Capabilities

Beyond Batch: Integrated & Continuous Chromatography

The Contichrom® CUBE doesn’t just run Batch — it’s engineered for the multi-column continuous processes that fundamentally improve purification economics. CaptureSMB® for capture, MCSGP for polishing, N-Rich® for impurity enrichment, and 2D/3D integrated batch — all powered by patented dynamic process control (AutoPeak® & AutomAb®) and accessible from a single platform.

2D/3D Integrated Batch

Link two or three orthogonal purification steps into a single automated process. Product transfers directly from column 1 to column 2 (and optionally column 3) with in-line dilution — no hold tanks, no manual handling.

Example Workflow Steps
mAb 2-step Protein A → Desalting
mAb 3-step Protein A → CIEX → Desalting
TIDES 2-step RP polishing → AIEX polishing

MCSGP with AutoPeak® — Continuous Polishing

Twin-column continuous polishing that eliminates the purity-yield trade-off. MCSGP internally recycles low-purity side-fractions between columns with in-line dilution. AutoPeak® monitors elution in real-time and dynamically adjusts collection windows to compensate for retention shifts, column aging, and feed variability.

  • Target: Gradient polishing separations
  • Molecules: Oligonucleotides, peptides, mAbs, proteins, AAV, mRNA

MCSGP with AutoPeak® — Performance Gains

Metric Result
Yield increase +10–50% absolute at target purity
Throughput Up to 5× higher
Re-chromatography Eliminated
Column size Smaller — parallel processing removes idle time
Process control AutoPeak® enables unattended 24/7 operation

One column elutes while the other loads — continuous output, zero downtime, consistent quality.

CaptureSMB® with AutomAb® — Continuous Capture

Twin-column continuous capture that maximizes resin utilization. CaptureSMB® loads columns beyond breakthrough in a countercurrent fashion — breakthrough from column 1 is captured on column 2. AutomAb® monitors the breakthrough curve via in-line UV and dynamically adjusts loading duration to compensate for feed titer variations and column aging.

  • Target: Affinity capture steps (Protein A)
  • Molecules: mAbs, bi-specifics, mRNA, vaccines, recombinant proteins

CaptureSMB® with AutomAb® — Performance Gains

Metric Result
Productivity 2–4× higher
Resin volume 20–50% reduction
Buffer consumption Up to 50% reduction
Resin binding capacity used >90% vs. 50–70% in batch
Process control AutomAb® adapts loading to titer variability in real-time

Smaller columns, smaller buffer tanks, reduced manufacturing footprint.

N-Rich® with AutoPeak® — Impurity Enrichment

Automated twin-column cyclic process for isolating low-abundance impurities or variants. N-Rich® accumulates target compounds on-column over multiple cycles, then elutes in a final high-resolution step. AutoPeak® compensates for retention time shifts, enabling robust unattended operation across many enrichment cycles.

  • Target: ICH-required impurity characterization
  • Molecules: Impurities from oligonucleotides, peptides, mAbs, proteins, AAV, mRNA

N-Rich® with AutoPeak® — Performance Gains

Metric Result
Processing speed Up to 80× faster (peptide)
Timeline 32 days → 3 days (mAb impurity isolation)
Purity improvement 11× over semi-prep batch
Solvent consumption Up to 70× reduction
Process control AutoPeak® delivers consistent purity across all cycles

Four stages: Design → Enrichment → Depletion → Elution.

Batch Chromatography

The CUBE handles single-column batch purification methods with full gradient and isocratic capability:

Mode Applications
Isocratic Protein A/L/G capture, SEC, desalting
Gradient IEX, RP, mixed mode polishing

100-bar design supports resins down to 10 µm. Fraction collection, UV/conductivity/pH monitoring, and ChromaCraft™ method design included as standard.

Alternating Batch

Run two columns in sequence on a single system — while one column elutes, the other equilibrates or loads. Alternating batch eliminates idle time between runs, effectively doubling sample throughput without any continuous process complexity.

  • Supported in: ChromaCraft™ Batch Editor
  • Use case: High-throughput screening, column comparisons, or maximizing daily output from routine purifications
